Question

When a hot 20-something colleague comments about the “hot 30- and 40-year olds” to me (40-something), is it an idea they’re trying to plant in my head, that they’re game for someone older like me?

No man, it doesn't.
It's just a fact that a lot of people (more women than men) look good for their age due to either good genetics or upkeep or both.

OP: gay or lesbian,go for it.
OP:if you are a woman and it's a younger guy then go ahead as you'll be applauded and be the envy of your female colleagues.
OP:if you are a man,ignore this younger woman. Perhaps you'll go out on a date and she won't like you,or maybe you'll flirt back and then you'll be hauled up to HR for sexual harassment,be fired and labelled a creepy pervert. Then, be unable to find work for a very long time.

A 40 something who doesn't know how to navigate this isn't secure enough to be trusted with the well being of a 20 something.

This is at work. Keep it in your pants until you can be certain you're not making her workplace uncomfortable with your supposed interest.
